RESEARCH FOCUS

  • Processes that control the movement of the phytohormones auxin, brassinosteroids, and ethylene precursors to regulate programmed and plastic plant development
  • Mechanistic function of ATP Binding Cassette (ABC) transporters
  • Dynamics of substrate interactions with transport proteins and their lipid environment
  • Integration of light and gravity responses and modification of auxin fluxes  by environmental stress.
  • Interactions of plant defence and developmental signalling

Biography

Biography:

Research in the lab is focused on the processes that control the movement of auxin and brassinosteroid phytohormones to regulate programmed and plastic plant development.  Much of the work focuses on how those processes respond to environmental inputs and stress. Current efforts involve integration of light and gravity responses and modification of auxin fluxes by biotic and abiotic stressors. The lab also studies the transport of other aromatic acids that function in plant / fungal growth processes. We use the tools of structural biology, modeling, biochemistry, cell biology, comparative systems biology, and molecular genetics in our work.

Plant hormone transport and growth

Plant hormone transport and growth

One of the longest standing projects in the lab investigates the cellular mechanisms that regulate auxin transport in dicots (Arabidopsis, tomato, cucurbits) and monocots (rice, maize, sorghum). This work has included mechanistic studies of auxin transporters and their regulation, differentiation of long distance and localized vectorial auxin transport streams, and the contributions of these streams to plant architecture and plastic responses to light and nutrients. Over the past two decades, the lab has played a major role in identifying and characterizing the auxin transport proteins from the ATP-Binding Cassette class B (ABCB) family, characterizing the subcellular mechanisms that regulate their activity, and clarifying their interactions with the PIN family of auxin transporters. More recent work has focused on light / gravity regulation of auxin transport and the impacts of salt stress on these processes. Funding for this project has come from the National Science Foundation, the Department of Energy, the US Department of Agriculture, and the Biotechnology and Biological Sciences Research Council (UK).

Structure-function analyses of ABCB transporters

Structure-function analyses of ABCB transporters

b focuses on elucidation of the structural features of plant ABCB transporters that confer the relative substrate specificity as well as some conditional uptake.  A major focus of this work is the generation of new ABC transporters to enhance production of engineered biofuel precursors. This work is funded by the Basic Energy Sciences Division of the  U.S Department of Energy.

herbicide uptake in plants and mammals

Herbicide uptake in plants and mammals

Movement of environmentally-applied herbicides into non target sites is a major public concern. This project in the lab was originally directed to understanding of the extent to which herbicides “drift” onto non-target crops and ornamentals, but, more recently, has also examined movement of herbicides into pets and households.
